Spider-Man: Brand New Day was primarily filmed in the United Kingdom, despite its New York City setting. The majority of the production took place at Pinewood Studios in Buckinghamshire, with digital visual effects and various location shoots across Scotland and England used to recreate iconic American scenes.

Glasgow, Scotland served as a stand-in for Manhattan in several scenes, including a high-speed chase involving a hijacked tank. Streets like Bothwell Street, St. Vincent Street, Hutcheson Street, and Wilson Street were used, with local businesses such as Chaakoo Bombay Cafe remaining open for visitors during production.

Basingstoke and Hertfordshire provided set pieces for the Damage Control Westside Branch facility, with Fanum House and Hertfordshire County Hall respectively. These locations were used for scenes featuring Peter Parker posing as a student at Empire State University to discuss his spider DNA with Professor Bruce Banner.

London landmarks, such as Senate House in Bloomsbury, Ravenscourt Park in Hammersmith, and West Carriage Drive in Battersea Park, were digitally enhanced to become the central headquarters for Damage Control, Empire State University, and various outdoor scenes featuring Tom Holland and Zendaya.

For Frank Castle's boathouse hideout, Turk's Shipyard within the Historic Dockyard Chatham in Kent was utilized. Additionally, scenes between Spider-Man and the Hand were filmed at the Dorchester Prison in Dorset, a decommissioned 19th-century jail. Finally, emotional scenes involving Peter visiting Aunt May's grave were captured at Brookwood Cemetery in Surrey, the UK's largest burial ground.
