Protein palmitoylation, a reversible lipid modification with significant implications in disease, remains poorly annotated in the scientific community. Existing databases heavily rely on isolated literature findings and lack standardized harmonization across mass spectrometry-based palmitoylome datasets, hindering comprehensive analyses.
To address this, we present PalmLab, a computational platform that integrates MS-based palmitoylome data curation with interactive, multi-dimensional analytical tools for both human and mouse proteomes. By employing a unified, rigorously standardized bioinformatics pipeline, PalmLab enables systematic reanalysis of all published palmitoylome datasets, resulting in a curated collection of 15,968 human and 9,924 mouse MS-validated palmitoylated proteins – a more than doubling of entries compared to existing public repositories.
In addition to its annotation capabilities, PalmLab offers user-friendly, code-free analytical modules for differential palmitoylation profiling, visualization of context-specific patterns, inference of protein-protein correlation networks, mapping of mutation-palmitoylation associations, and discovery of sequence-based motifs. As a freely accessible resource, PalmLab provides a comprehensive platform for systematic mining and functional interrogation of palmitoylation events, paving the way for elucidating palmitoylation-mediated regulatory circuits and prioritizing novel therapeutic targets in oncology.
