"account": "Parathyroid hormone (PTH) plays a crucial role in regulating bone remodeling by coupling osteoclast-mediated bone resorption with osteoblast-mediated bone formation. The precise timing of PTH exposure influences the dynamics of this process, but the impact of exposure frequency on spatiotemporal coupling during bone remodeling is not well understood. To investigate this, researchers utilized a functional bone model (FBM) composed of osteoblasts and osteoclast precursors. The FBM encompasses formation, resorption, and reformation phases, and PTH(1-34) was administered during the resorption and reformation phases. Using longitudinal two-photon microscopy with second-harmonic generation imaging, the study tracked matrix resorption and reformation, quantifying spatiotemporal changes in cell volume and morphology. Seven regimens were evaluated: vehicle, intermittent PTH once or three times weekly at 10 or 50 nM, or continuous PTH at 10 or 50 nM. The findings revealed that once-weekly intermittent PTH enhanced osteoclastogenesis and resorption while maintaining spatial coupling, osteoblast repopulation, and recovery of resorption volume. Conversely, three-times-weekly and continuous PTH exposure led to more extensive matrix loss, weaker spatial coupling between resorption areas and reformation, and limited reformation. These results highlight that exposure frequency, in conjunction with dose, is a critical determinant of PTH-driven remodeling trajectories. The study demonstrates that PTH(1-34) exposure frequency can tune coupling in the FBM, suggesting the potential of this model for optimizing PTH-based regenerative strategies and advancing broader bone remodeling research.",
